We investigated 296 adolescents (11–18 years), who had been immunized previously with the three doses of DPT vaccines. 48 were diagnosed positive for HIV-1. Nasopharyngeal swabs were obtained from 296 adolescents who presented with persistent cough and nasopharyngeal secretions. Nasopharyngeal swabs (calcium alginate) specimens were collected by passing the swabs through the nares into the posterior nasopharynx and rotating the swabs for a few seconds. The swabs were plated for culture of Bordetella organisms in charcoal cephalexin blood agar (CCBA). The CCBA plates were incubated for 2–6 days at 35 °C in a humid aerobic atmosphere. The suspected, shiny (mercury-like) colonies were tested by slide agglutination with antisera to B. pertussis and B. parapertussis, and urease, oxidase activities were performed. Results indicate that out of 48 HIV-1-positive adolescents, 18 had positive cultures for Bordetella organisms (14, Bordetella pertussis, and 4, Bordetella parapertussis). Of 248 HIV-1-negative subjects, 3 had Bordetella organisms (2, Bordetella pertussis, 1, Bordetella bronchiseptica). One of the subjects, a boy, aged 14 years, with Bordetella bronchiseptica had a dog as pet, which was found to be infected. The results indicate that adolescents with HIV-1 infection, despite being vaccinated against pertussis have a higher rate of infection when exposed to pertussis bacteria than HIV-1-negative adolescents. Smoke, canopy‐derived mulch, and broadcast seeds were used to maximize the establishment of Banksia woodland species in sand quarries in Western Australia. Smoke, particularly aerosol smoke, had a positive effect on total seedling recruitment. Pre‐mined (woodland) sites showed a 42‐fold increase in total germinants and a 3‐fold increase in the number of species with aerosol smoke application. Post‐mined (restored) sites showed only a 3.6‐fold increase in total germinants and a 1.4‐fold increase in the number of species. Two water‐based smoke chemicals, DC10 (pH 4.5) and SC63 (pH 2.5), increased seedling recruitment at both sites but were not as effective in stimulating recruitment as aerosol smoke. Neither of the chemicals were effective in significantly increasing species richness. Application of aerosol smoke directly to seeds as a pretreatment before broadcasting had no effect on seedling recruitment. Broadcasting of seeds onto restoration sites significantly increased seedling abundance and richness. Application of a single layer of mulch from the canopy vegetation after seed broadcasting gave optimum seedling recruitment. Two layers of mulch significantly reduced recruitment, as did applying mulch before seed broadcasting. For broad‐scale restoration, the application of smoke on newly restored sites would be more effectively achieved using smoke water sprayed over the soil surface. Species that do not recruit from replaced topsoil could be effectively recovered from broadcast seed rather than from mulch.  相似文献

The most common disease syndromes caused by Salmonella serotypes in humans, typhoid fever and enteritis, can be modeled using Salmonella enterica serotype Typhimurium infections in mice and calves, respectively. Murine Bmp7 has been assigned to distal Chromosome 2 by interspecific backcross mapping. The map location suggests close linkage to classical mouse mutations and places Bmp7 within a chromosome region thought to contain one or more unidentified imprinted genes. A direct test suggests that Bmp7 is not imprinted. An examination of embryonic RNA expression patterns shows that Bmp7 is expressed in a variety of skeletal and nonskeletal tissues. Both embryonic expression patterns and the human chromosomal sublocalization inferred from its mouse location make Bmp7 a candidate for the gene affected in some patients with Holt-Oram syndrome.  相似文献

Segments of a cDNA encoding human plasminogen activator inhibitor type 1 (PAI-1) were subcloned into a highly regulated and inducible Escherichia coli expression system. A plasmid encoding the mature form of human endothelial PAI-1 produced a functional recombinant molecule, as indicated by its ability to inhibit tissue plasminogen activator's enzymatic activity. In contrast to PAI-1 isolated from human fibrosarcoma cells, the biological activity of the recombinant PAI-1 was not dependent on pretreatment with denaturing agents. A construct encoding a polypeptide lacking the first 80 amino acids of PAI-1 also produced elevated levels of the truncated recombinant protein. However, this truncated product was functionally inactive, indicating that an intact N terminus is required for activity.  相似文献

Aim The causes of orchid diversification and intrinsic rarity are poorly resolved. The Orchidaceae of the Southwest Australian Floristic Region use a diversity of pollination strategies and sites of mycorrhizal infection, and occupy a diversity of habitats. We combined a biogeographic analysis with analysis of factors associated with rarity to establish: (1) the landscape features correlated with taxon turnover and speciation, and (2) the possible role in taxon rarity of geographic region, pollination strategy, edaphic habitat and site of mycorrhizal infection. Location Southwest Australian Floristic Region. Methods The distributions of 407 orchid taxa (species and subspecies) were mapped at the quarter‐degree scale using 13,267 collections in the Western Australian Herbarium. This database was used to map taxon richness, for a biogeographic analysis and to quantify rarity of taxa. Using herbarium records, rarity was expressed as mean abundance, mean distribution and incidence of rarity based on abundance and distribution for each genus. We tested for differences in rarity of species between pollination strategies, edaphic habitats and sites of mycorrhizal infection. Results Taxon richness was highest in the High Rainfall Province. Biogeographic provincial boundaries for orchids were aligned with rainfall, while district boundaries tended to follow geological formations. When rarity was defined as either low abundance or small distribution, the greatest number of rare taxa occurred in areas of high taxon richness and naturally fragmented edaphic environments. For both abundance and distributional extent, sexual deception had a significantly higher incidence of rarity than food‐rewarding taxa. There was no significant difference in rarity with site of mycorrhizal infection. Main conclusions While large‐scale edaphic and climatic variation are correlated with orchid taxon turnover and speciation in a similar fashion to the flora in general, the processes responsible for patterns of diversity may differ. Fragmented edaphic environments appear to be associated with a higher incidence of rare species due to limited dispersal/colonization opportunities or radiations of taxa in allopatry. The high incidence of rarity in sexually deceptive taxa could be due to either low fruit set or the risk of specializing on a single pollinator species.  相似文献

In response to a critique by Higgs et al., this article clarifies the content and intent of the Society for Ecological Restoration's (SER) International Standards for the Practice of Ecological Restoration. Higgs et al. expressed concern that the SER Standards are not sufficiently underpinned by principles and risk disenfranchising some practitioners by narrowing what qualifies as ecological restoration. To demonstrate that these concerns are unfounded, we discuss the policy context and principles on which the Standards are based, its organizational structure, the innovative and inclusive approach used for development, and highlight significant errata by Higgs et al.  相似文献

Species distribution models can be informative of the biodiversity impacts of changing environments at global, national, and regional scales, but are often constrained in their resolution to extents not relevant to individual, intensive ecological management programs. We constructed a high‐resolution topoclimatic model of spring and summer temperatures across a 152 km2 restoration area on the Swan Coastal Plain, Western Australia, and used it to project energetic expenditure and habitat suitability estimates for four major hymenopteran pollinators. For all species, the most heavily modified landscapes were the least suitable, but only for one species, Zapsilothynnus nigripes, was there evidence that the upper thermal tolerance threshold was exceeded broadly. However, at the higher environmental temperatures that we modeled, some species would need to forage up to 10 times their own body mass every hour to meet their energetic requirements. It seems unlikely that the nutritional requirements of most insect pollinators operating at these higher metabolic rates could be met in an impoverished restoration ecosystem, although resource availability remains to be quantified in these habitats. Hence, to increase the likelihood of restoration success by restoring insect pollination networks, nutritional resources may need to be increased during restoration. Accounting for the way that thermoenergetic requirements shape ecological interactions better positions management trajectories aimed at restoring and maintaining key insect pollinators in “novel” ecosystems.  相似文献
